At the recent American Society of Clinical Oncology (ASCO) Annual Meeting, CStone Pharmaceuticals presented new clinical data that sheds light on its innovative trispecific antibody, CS2009, which targets PD-1, VEGF, and CTLA-4. This groundbreaking agent demonstrates significant potential to transform the landscape of cancer immunotherapy, particularly in treating both non-small cell lung cancer (NSCLC) and colorectal cancer (CRC).
CS2009 in First-line NSCLC Treatment
A standout finding from CStone’s study was the remarkable effectiveness of CS2009 in first-line NSCLC patients expressing high PD-L1 levels (TPS ≥50%). In this cohort, CS2009 as a monotherapy achieved an objective response rate (ORR) of 81.3% and a stunning disease control rate (DCR) of 100%. These effects were consistent across both squamous (ORR 87.5%) and non-squamous (ORR 75%) histologies. Adding to the excitement was the performance of CS2009 in PD-L1-negative/low patients, where a combination of CS2009 with chemotherapy yielded an ORR of 75% and a DCR of 100%, featuring a 100% ORR among the sub-group of patients with a TPS <1%.
Overcoming Treatment Resistance in Later-line NSCLC
In patients with heavily pretreated later-line NSCLC, CS2009 showcased promising antitumor activity, with most participants experiencing tumor shrinkage. Notably, across all dose levels, the six-month duration of response rate reached an impressive 85.7%. In a combination cohort involving second- and third-line treatment, CS2009 boasted an ORR of 66.7% and a DCR of 100%. Furthermore, CS2009 monotherapy in those whose disease progressed after prior immunotherapy achieved an ORR of 30.8%, highlighting its potential as a remedy for treatment-resistant cases.
