ArkBio Receives FDA Approval for Phase II Trial of AK0406 to Combat Influenza

ArkBio Receives FDA Clearance for Phase II Trial of AK0406

In an important milestone for influenza prevention, Shanghai Ark Biopharmaceutical Co., Ltd. — known as ArkBio — has received approval from the U.S. Food and Drug Administration (FDA) to initiate a Phase II clinical trial of its investigational new drug, AK0406. This long-acting antiviral drug-Fc conjugate (AFC) is designed to provide robust defense against influenza and is poised to make a notable impact in public health.

Addressing Challenges in Influenza Prevention

Influenza remains a significant public health concern, with seasonal vaccinations being a cornerstone of prevention efforts. However, variability in virus strains and individual immune responses can diminish the efficacy of these vaccines, particularly among vulnerable populations such as older adults and those with weakened immune systems. Despite the availability of vaccines, the CDC reported a staggering number of affected individuals — at least 32 million illnesses, 380,000 hospitalizations, and 24,000 deaths during the 2025–2026 influenza season alone.

About AK0406

AK0406 represents a novel approach in antiviral therapy, developed by ArkBio through precise conjugation of multiple potent small-molecular antivirals to an antibody Fc domain. This unique formulation not only targets the virus directly but also enhances immune clearance — a dual action aimed at preventing influenza more effectively. In preclinical studies, AK0406 exhibited broad-spectrum antiviral activity against both influenza A and B strains, showcasing promising potential for future therapeutic applications.

Global Clinical Development Journey

Prior to the FDA's clearance, AK0406 had garnered attention globally. In February 2026, it gained approval from a Human Research Ethics Committee (HREC) in Australia to commence a Phase I trial, which successfully dosed healthy adult participants across all cohorts by July 2026. ARK has also received support from the National Medical Products Administration (NMPA) of China, making significant progress in its clinical development across multiple regions.

Planned U.S. Phase II Trial

The upcoming Phase II clinical trial in the United States will be a multicenter, randomized, double-blind, placebo-controlled study targeting adults during the influenza season. The study aims to comprehensively evaluate AK0406 for its efficacy in preventing influenza infections while also assessing its safety, tolerability, pharmacokinetics (PK), and immunogenicity. Insights gained from this trial will be instrumental in shaping the future global development strategy for AK0406.

ArkBio is committed to working closely with the FDA and will continue to dialogue with the CDE of China’s NMPA to advance this promising product's development in both nations.

About ArkBio

Founded in 2014, ArkBio is focused on pioneering the discovery and development of advanced therapeutics for respiratory and pediatric health issues. Their research and development pipeline is robust, featuring other promising drugs such as ziresovir (AK0529), which has shown success in Phase III trials for RSV, and AK3280, an anti-fibrotic agent currently undergoing Phase III trials for idiopathic pulmonary fibrosis. ArkBio illustrates the potential of biopharmaceutical innovations to enhance global health outcomes, particularly amid rising public health challenges like influenza.
