Xentria's Commitment to Address Pulmonary Sarcoidosis
Xentria, Inc., a biotherapeutics company focused on innovative treatments, has outlined its next steps in the clinical development of
XTMAB-16, an investigational treatment designed for pulmonary sarcoidosis. This rare condition currently lacks an approved
anti-TNFα therapy from the U.S. Food and Drug Administration (FDA), highlighting the urgent need for effective therapeutic options.
The XAtlas Study: Expanding the Clinical Horizon
On September 1, 2026, Xentria announced its intentions to amend the ongoing
XTMAB-16-201 clinical trial. In response to valuable feedback from the FDA following an end-of-phase meeting, the company plans to enhance the study design. The proposed
XAtlas study aims to broaden the enrollment from 62 to as many as 200 participants and extend the study duration to 52 weeks. Patients will receive
4mg/kg of XTMAB-16 once every four weeks, with the primary efficacy endpoint focusing on changes in forced vital capacity (FVC) from baseline. Additional measures will also be taken to assess disease management through several patient-reported outcomes.
Previous Study Findings: Safety and Efficacy
The XTMAB-16-201 Part A study, which was a randomized, placebo-controlled trial involving 39 adults across the United States and Europe, concluded successfully with 38 patients completing the regimen. Participants were administered XTMAB-16 at various doses over a 12-week trial period. The trial primarily focused on safety, tolerability, pharmacokinetics, and immunogenicity.
The results indicated that XTMAB-16 was well tolerated, with no significant treatment-emergent adverse effects or dose-limiting toxicities reported. No serious adverse events or fatalities were associated with the treatment, though it's essential to acknowledge that the limited sample size means comprehensive safety profiles for XTMAB-16 are still under investigation. The exploratory phase of the trial suggested positive trends in managing corticosteroid use and preserving lung function.
Significance of Addressing Pulmonary Sarcoidosis
Pulmonary sarcoidosis poses a serious health threat, leading to chronic, unresolved inflammation that can result in lung scarring and impaired function. This disease affects more than 90% of the 1-2 million individuals with sarcoidosis worldwide, creating an urgent requirement for effective therapies. While the exact cause remains elusive, the role of
TNFα in driving inflammatory processes in sarcoidosis emphasizes the potential therapeutic value of XTMAB-16, which aims to disrupt inflammatory pathways and mitigate the formation of granulomas.
Future Outlook for Xentria and XTMAB-16
Xentria's CEO,
Tom Shea, highlights the importance of the FDA's feedback as instrumental in shaping the upcoming clinical development protocol. The XAtlas study's operational plans are set for 2026, reflecting Xentria's commitment to addressing the pressing needs of patients with pulmonary sarcoidosis. By implementing insights from regulatory authorities, Xentria seeks to push forward with the exploration of XTMAB-16 as a novel treatment option.
