Innovent Biologics Unveils Promising Phase 2 Data for IBI354 in Breast Cancer Treatment at ESMO 2026

Innovent Biologics' Presentation at ESMO 2026

On October 25, 2026, Innovent Biologics, Inc. delivered a pivotal announcement during the European Society for Medical Oncology (ESMO) Congress regarding its Phase 2 clinical trial data for IBI354, an innovative HER2-targeted antibody-drug conjugate (ADC) aimed at treating HER2-positive breast cancer as a first-line therapy. This landmark presentation underscores Innovent's commitment to addressing significant health challenges through groundbreaking biopharmaceutical advancements.

Overview of IBI354

The HER2 ADC, IBI354, is part of Innovent’s extensive efforts to develop effective treatments for difficult-to-treat cancers and other major diseases. As part of the comprehensive clinical development strategy, IBI354 is designed to maximize anti-tumor activity while minimizing toxicity. With its promising safety profile, IBI354 stands as a potential leading treatment option in oncology, capable of changing the landscape of HER2-positive breast cancer care.

Recent data showed a confirmed objective response rate (cORR) of 95% and a disease control rate (DCR) of 100% in patients receiving IBI354 in conjunction with pertuzumab. These striking figures highlight the promising efficacy of this therapy where conventional treatments have struggled.

Efficacy and Safety Profile

In the study, 20 patients suffering from HER2-positive unresectable locally advanced or metastatic breast cancer were treated with IBI354 at a dosage of 9 mg/kg every three weeks, in combination with pertuzumab. The median follow-up duration for this study was 9.6 months, with treatment generally well-tolerated. Notably, the tenacity of the treatment against the malignancy showcased through a median progression-free survival (PFS) of 8.3 months was encouraging, although final efficacy data remains pending.

The safety profile of IBI354 is also noteworthy. The most frequent treatment-emergent adverse events (TEAEs) included nausea, vomiting, and anemia; however, no cases of severe interstitial lung disease (ILD) were reported, which is a common concern with other ADC treatments. Only 20% of patients experienced grade 3 or higher treatment-related adverse events, emphasizing the favorable safety compared to historical benchmarks in the field.

Expert Opinions

Professor Xiaohua Zeng from Chongqing University Cancer Hospital emphasized, “As research into HER2-positive advanced breast cancer progresses, the introduction of innovative therapies such as IBI354 marks a significant improvement in patient management and treatment success.” He raised important points regarding the need for ongoing optimization of treatment safety and efficacy to further enhance patient outcomes.

Dr. Hui Zhou, the Chief R&D Officer for Innovent Biologics, reaffirmed the company's dedication to advancing cancer treatment through ongoing innovation. He stated, “The data presented reinforce our commitment to providing patients with highly effective and safe treatment options. We are looking forward to expanding our ADC pipeline and further developing IBI354.”

Future Directions

Following the promising Phase 2 results presented at ESMO 2026, Innovent has initiated pivotal Phase 3 studies (HeriCare-Ovarian01 and HeriCare-Breast01) aimed at validating the effectiveness of IBI354 across a broader spectrum of patients and possibly initiating studies for other tumor types, including ovarian cancer. Innovent’s broad pipeline aims to ensure patients have access to the most advanced, effective therapies available.

Innovent Bio, founded in 2011, maintains a mission aimed at delivering affordable, high-quality biopharmaceuticals to empower patients globally. With more than 20 products already on the market, and ongoing trials aimed at addressing pressing medical needs, Innovent is positioned at the forefront of oncology research.
