Sun Pharma to Present New Data on UNLOXCYT® in Advanced Skin Cancer at ESMO 2026

Sun Pharmaceutical Industries Limited is set to reveal significant long-term follow-up data regarding UNLOXCYT® (cosibelimab-ipdl) during the European Society for Medical Oncology (ESMO) Congress on October 24, 2026, taking place in Madrid, Spain. This data emerges from one of the largest prospective trials concerning advanced cutaneous squamous cell carcinoma (aCSCC), specifically targeting locally advanced cutaneous squamous cell carcinoma (laCSCC) and metastatic cutaneous squamous cell carcinoma (mCSCC).

UNLOXCYT® was granted approval by the U.S. Food and Drug Administration (FDA) in 2024, designed for adult patients with aCSCC who are not suitable candidates for curative surgery or radiation therapy. The recommended dosage is 1,200 mg administered as an intravenous infusion over 60 minutes every three weeks. Notably, it stands as the inaugural immune checkpoint inhibitor targeting aCSCC, aiding in the restoration of adaptive immune function while preserving PD-L2 signaling. This mechanism likely plays a role in maintaining immune tolerance within non-tumorous tissues, supporting the observed favorable tolerability profile.

In the trial, data was centrally reviewed across three groups totaling 205 patients with advanced CSCC. The first group, comprising mCSCC patients, received a dosage of 800 mg every two weeks; the second group included laCSCC patients with the same regimen, while the third group of mCSCC patients were treated with 1,200 mg every three weeks. The principle aim was to ascertain the objective response rate (ORR) by central review, with secondary focus on the duration of response (DOR), progression-free survival (PFS), overall survival (OS), and safety assessments.

Dr. Eva Muñoz-Couselo, a significant contributor to the study, emphasized the noteworthy durability of responses observed over time, alongside an increased number of patients achieving complete responses—a vital outcome for individuals grappling with this challenging disease. The results also displayed a persistently low incidence of severe immune-related adverse events (irAEs), reinforcing the drug's potential to deliver sustained disease control with manageable safety concerns. The overall response rates align with previous analyses, while additionally revealing an upward trend in complete responses.

Within the analyzed cohort, notable statistics emerged: In Group 1, the ORR was recorded at 51.3%, with a complete response (CR) rate of 16.7%. The median duration of response was an impressive 45.9 months, with a median follow-up duration of 49.9 months. Group 2 showcased an ORR of 53.1% and a CR rate of 34.4%, with a median follow-up of 36 months and an 81.8% response estimate at 24 months. Group 3 reported an ORR of 44.4% with a corresponding CR rate of 20.6% and a median follow-up of 29.9 months, evidencing a strong response persistence among individuals treated.

In a broader context, an estimated 40,000 individuals in the United States transition to advanced forms of cutaneous squamous cell carcinoma each year, leading to nearly 15,000 fatalities. Recognized risk factors are extensively linked to prolonged exposure to ultraviolet radiation and immunosuppressive conditions. Beyond their life-threatening nature, these tumors frequently inflict notable functional disabilities and aesthetic disfigurements, especially those arising in regions prone to visibility, such as the head and neck.

As part of its ongoing commitment to enhancing treatment access, Sun Pharma continues to explore avenues for expanding the availability of UNLOXCYT to healthcare providers and patients outside the U.S. The company has been at the forefront of dermatological care for over two decades, focusing on the myriad treatment needs across the disease spectrum from precancerous conditions to advanced skin cancers.

In summary, the upcoming data presentation at ESMO 2026 promises to deliver critical insights into the efficacy and safety of UNLOXCYT, highlighting its significant role in managing advanced cutaneous squamous cell carcinoma. This milestone reflects Sun Pharma's dedication to addressing the unmet needs of patients afflicted by this often-neglected but serious form of skin cancer. As research progresses, the insights gained may pave the way for enhanced treatment modalities and improved patient outcomes in the future.
