New Phase 3 Data Reveals Efficacy of Pozdeutinurad in Reducing Uric Acid in Gout Patients

The recent late-breaking data from the Phase 3 REDUCE-2 trial has garnered attention in the medical community, particularly in the field of rheumatology. Conducted by Sobi®, the study evaluated the investigational drug pozdeutinurad (AR882), a highly selective URAT1 inhibitor designed for patients suffering from uncontrolled gout. This pivotal research was presented at the ACR Convergence 2026, showcasing statistically significant reductions in serum urate (SU) levels.

In the study, a total of 811 participants with gout were involved. They were randomly assigned to receive either a 75 mg dose, a 50 mg dose of pozdeutinurad, or a placebo. The results were striking; after six months, 69.2% of patients taking the 75 mg dose and 56.6% of those on the 50 mg dose achieved SU levels below 6 mg/dL, compared to a mere 8.1% of those receiving the placebo (both p<0.0001). It’s remarkable that these reductions in serum urate began as early as one month into treatment and were sustained through the entire 12-month duration of the study. Throughout this period, patients receiving pozdeutinurad consistently demonstrated superior results in achieving lower SU thresholds (6, 5, 4, and 3 mg/dL) compared to placebo groups.

The safety profile of pozdeutinurad was also closely monitored. Notably, the incidence of treatment-emergent adverse events (TEAEs) was similar across all groups, occurring in approximately 60% of participants regardless of whether they received the drug or the placebo. This consistency suggests that pozdeutinurad is generally well-tolerated and presents a favorable safety profile when compared to existing therapies.

Dr. Puja P. Khanna, a prominent researcher from the University of Michigan and investigator in the REDUCE-2 trial, commented on the findings. According to Dr. Khanna, achieving and maintaining target serum urate levels is crucial for effective gout management. Many patients find themselves inadequately treated with current oral therapies, which can precipitate irreversible joint and tissue damage. The evidence provided by the REDUCE-2 study illustrates that pozdeutinurad can lead to meaningful and sustained serum urate lowering, reinforcing its potential as an innovative treatment option for those grappling with uncontrolled gout.

Dr. Lydia Abad-Franch, Sobi’s Chief Medical Officer, expressed excitement over the implications of these late-breaking findings. She emphasized the importance of these results for innovation in gout therapies, especially noting the encouraging consistency across different dosages of pozdeutinurad. As the company continues to explore its benefits, the medical community may soon gain access to an important tool for managing this challenging condition.

To better understand gout, it’s essential to recognize that it is the most prevalent form of systemic inflammatory arthritis. The condition arises from elevated serum urate levels leading to the accumulation of urate crystals in the joints and other tissues, resulting in intense pain and potential joint damage. Uncontrolled gout, characterized by serum urate levels exceeding 6 mg/dL, presents ongoing challenges for patients, including painful flares and tophi that persist despite current treatments. The progressive nature of this condition is underscored by its links to various other diseases, such as cardiovascular and renal disorders. Thus, sustaining a lower serum urate level is critical for effective gout management, facilitating the dissolution of urate crystals and alleviating symptoms.

Pozdeutinurad’s clinical development is part of a larger investigation involving over 800 patients within two Phase 3 studies - REDUCE-1 and REDUCE-2. The drug aims to present an effective oral solution that not only accomplishes significant SU reduction but also addresses the unmet needs of patients struggling with uncontrolled gout. As interest in this new treatment option grows, it could reshape the therapeutic landscape for individuals affected by gout, providing both relief and improved quality of life.

This pivotal research heralds a vital step in advancing treatment options for gout, a condition often misunderstood but affecting countless individuals worldwide. The implications of pozdeutinurad could resonate far beyond the trial results, offering hope to patients who have long awaited meaningful advancements in gout therapy.
