AbelZeta Publishes Groundbreaking CAR-T Therapy Findings in Nature Journal

Background of C-CAR031
C-CAR031 is the result of a strategic collaboration between AbelZeta and AstraZeneca, specifically designed to leverage the body's immune response against cancers. The therapy employs an advanced armoring technology known as the dominant-negative transforming growth factor-beta receptor II (dnTGFβRII), which is instrumental in overcoming the immunosuppressive tumor microenvironment prevalent in HCC cases. This capability aims to enhance the efficacy of the CAR-T cells in targeting tumor cells more effectively.
In the groundbreaking trial led by Dr. Tingbo Liang and Dr. Qi Zhang from Zhejiang University School of Medicine, C-CAR031 was tested on 36 patients who had already undergone extensive treatment regimens, averaging four lines of prior therapy. The findings indicate promising results, showcasing a notable degree of tumor regression in a majority of cases. Out of the 36 patients, 32 exhibited tumor regression, with the median reduction being an impressive 41.6% from baseline.
Efficacy and Safety Results
According to the data, the objective response rate reached 44.4%, with a median duration of response documented at 4.4 months. Notably, the median progression-free survival (PFS) time stood at 4.2 months, while median overall survival (OS) extended to 14.2 months, underlining the potential durability of responses in this challenging patient cohort.
While the results are encouraging, the safety profile of C-CAR031 was also meticulously reviewed. Two patients experienced grade 3 cytokine release syndrome, while nine faced non-hematological adverse events of grade 3 or higher. These findings accentuate the necessity for continued monitoring as the therapy progresses through its clinical stages.
