Innovent Presents Updated Phase 1 Clinical Data for IBI3003 in Myeloma Patients



Innovent Biologics, Inc., a leading biopharmaceutical firm, has recently made significant announcements regarding the updated Phase 1 clinical data for its innovative trispecific antibody, IBI3003. This novel therapy targets GPRC5D, BCMA, and CD3 and is designed for patients suffering from relapsed or refractory multiple myeloma (R/R MM). The update was shared during an oral presentation at the 23rd International Myeloma Society (IMS) Annual Meeting 2026.

Key Findings from the Phase 1 Trial



The results from the Phase 1 dose optimization study revealed that the recommended Phase 2 dose (RP2D) for IBI3003 was determined to be 360 μg/kg. At this dosage, the antibody exhibited rapid, profound, and sustained therapeutic responses, particularly in high-risk patients with extramedullary disease who had undergone extensive prior treatments. The data highlights IBI3003's dual-targeting mechanism, designed to prevent the tumor escape that can occur due to the loss or downregulation of a single tumor antigen.

In preclinical studies, IBI3003 outperformed existing bispecific antibodies targeting similar pathways, showcasing remarkable anti-tumor activity, especially in models with low expression of its targets. Currently, Innovent is conducting additional trials in various global locations, including China, Australia, and the United States, aimed at assessing the antibody's safety and initial efficacy.

Enrollment and Demographics



The Phase 1 study enrolled a total of 102 patients from China and Australia, all of whom had experienced failure with at least two prior treatment regimens, including proteasome inhibitors and immunomodulatory drugs. Notably, the cohort demonstrated diverse response rates, and the median age of participants was 62 years, with a significant portion (37.1%) classified as high-risk.

Efficacy and Safety Profile



The updated data from this study revealed an impressive overall response rate (ORR) of 84.6% at the RP2D of 360 μg/kg. Among the participants, 11 achieved stringent complete responses (sCR), while others noted very good partial responses (VGPR) and partial responses (PR). Additionally, a median time of 0.99 months was observed for the first response, indicating the swift action of the therapy.

Despite its potency, the safety profile of IBI3003 appeared manageable, with hematologic toxicities being the most commonly reported adverse events. Most were manageable and reversible, with mild incidences of cytokine release syndrome (CRS) noted in 52% of patients. Prophylactic measures were incorporated to mitigate CRS risks during treatment.

Pharmacodynamic Response and Biomarker Analysis



Analysis of biomarkers indicated a significant decline in serum soluble BCMA levels across all dosage cohorts, affirming the drug's sustained pharmacodynamic efficacy. These findings support the hypothesis that IBI3003 can potentially address the challenge of treatment resistance in heavily pretreated multiple myeloma patients.
