Acepodia's ACE723: Pioneering Dual-Payload Therapy in Phase 1 Trials

Acepodia's Milestone in Cancer Treatment: First Patient Dosed with ACE723

Acepodia, a biotechnology firm specializing in the development of advanced therapies for cancer, has marked a significant milestone by dosing its first patient in the Phase 1 clinical trial of ACE723. This groundbreaking therapeutic agent is designed to target and treat unresectable or metastatic hepatocellular carcinoma (HCC), one of the most common forms of liver cancer.

Underlying Technology

ACE723 is a dual-payload antibody-drug conjugate (ADC) that combines a GPC3-targeting antibody with two distinct cytotoxic agents. This innovative design aims to overcome the challenges associated with tumor heterogeneity and resistance that often complicate single-payload ADC therapies. GPC3, frequently expressed in HCC, serves as an optimal target for directly delivering cytotoxic payloads to tumor cells.

The trial itself represents the first clinical application of Acepodia’s proprietary Antibody-Dual-Drugs Conjugation (AD2C) platform. Dr. Sonny Hsiao, the CEO and Chairman of Acepodia, expressed gratitude towards patients, researchers, and clinical staff for their dedication to the project, emphasizing that the initiation of human trials is a crucial step for the company’s AD2C platform.

Clinical Trial Structure

The Phase 1 trial of ACE723 is set to evaluate several critical factors, including the safety profile, dose-limiting toxicities, pharmacokinetics, and preliminary anti-tumor activity. The trial is structured with a dose-escalation phase designed to ascertain a recommended dosage for future stages of development and potential expansion cohorts.

Acepodia received Investigational New Drug (IND) clearance from both the U.S. Food and Drug Administration (FDA) and China’s National Medical Products Administration (NMPA) in September 2026. Following this, the Taiwan Food and Drug Administration (TFDA) also granted approval in October, allowing the trial to progress across these three key regions: the U.S., China, and Taiwan.

The Urgent Need for New Treatments

With HCC accounting for over 70% of liver cancer cases worldwide, the demand for innovative treatments is more pressing than ever. The advancement of therapies such as ACE723 is particularly critical, as existing immunotherapy-based treatments often lead to progression and resistance, leaving patients with limited options for effective care. As such, dual-payload ADCs like ACE723 represent a promising avenue for addressing these unmet needs.

About Acepodia

Founded with the mission to develop next-generation therapies for cancer and autoimmune diseases, Acepodia employs revolutionary conjugation technology that incorporates bio-orthogonal click chemistry. This approach enables the company to merge antibodies with therapeutic agents or immune cells, creating versatile treatments tailored for various conditions, including solid tumors and hematologic malignancies.

Through its AD2C and Antibody-Cell Conjugation (ACC) platforms, Acepodia aims to harness the therapeutic efficacy of these modalities while ensuring their adaptability across a broad spectrum of disease states.

As clinical trials unfold, the journey of ACE723 will be closely monitored, representing not just a leap forward for Acepodia, but a potential breakthrough in the realm of cancer therapy.
