Impressive Progression-Free Survival Data for 3SBio's Bispecific Antibody at ESMO 2026

Groundbreaking Progression-Free Survival Results from 3SBio at ESMO 2026

At the recently concluded 2026 European Society for Medical Oncology (ESMO) Congress, 3SBio presented significant updates on its bispecific antibody, SSGJ-707, which combines PD-1 and VEGF targeting in advanced non-small cell lung cancer (NSCLC). The long-term follow-up data from a Phase II study highlighted impressive efficacy with a median progression-free survival (mPFS) reaching 13.6 months in squamous NSCLC (sq-NSCLC) and 13.8 months in non-squamous NSCLC (nsq-NSCLC). This advancement is a crucial marker in the development of effective treatments for lung cancer, showcasing the potential of innovative therapies from China.

Overview of the Clinical Study

The Phase II clinical study involved a diverse cohort of 261 patients – including 139 with sq-NSCLC and 122 with nsq-NSCLC. Among these patients, 50% were aged 65 or older, highlighting a critical demographic often underserved in cancer therapies. Moreover, the majority of participants had a notable history of smoking, and over 40% displayed a PD-L1 tumor proportion score (TPS) of less than one percent.

The regimen evaluated comprised tivunatamig administered in conjunction with platinum-based chemotherapy. In this setting, the confirmed objective response rate (cORR) for sq-NSCLC cases reached an outstanding 80.7%, compared to 67.7% in a control group, while nsq-NSCLC treated with the same regimen exhibited a cORR of 77.5% versus 44.7% for controls.

Continued Improvement in Efficacy

These results reflect a gradual enhancement in efficacy, a positive trend that physicians and researchers hope will continue into larger, Phase III trials. Notably, the latest follow-up data indicated that the overall treatment effect did not diminish over time, suggesting that the high response rates observed are genuine and not merely transient.

Moreover, the report from ESMO 2026 indicated lasting benefits that are crucial for physicians in evaluating long-term treatment plans. The uniqueness of this therapy was further underscored by its comparable safety profile to existing treatments, notably no new severe adverse events reported since previous disclosures. It is imperative for oncologists to have confidence in the safety of the treatment amid the promising efficacy.

ctDNA as a Potential Biomarker

One of the standout findings from this study was the analysis of circulating tumor DNA (ctDNA) in a subset of the patient population. Among patients with detectable baseline ctDNA, 61.9% experienced ctDNA clearance by the start of Cycle 3. This molecular indicator signifies a significant reduction in tumor burden, correlating with a median PFS of 16.4 months in the ctDNA-clearance group. In contrast, those without ctDNA clearance exhibited a significantly shorter PFS of 9.6 months.

This early observation points to a potential for ctDNA to serve as a predictive biomarker, enabling healthcare providers to identify which patients might derive substantial benefit from treatment sooner than traditional imaging techniques might allow.

A Competitive Landscape and Strategic Collaborations

The competitive landscape for advanced NSCLC treatments is intense, with multiple pharmaceutical entities vying for dominance. 3SBio’s partnership with Pfizer in 2025, which involved a substantial upfront payment for the global rights of SSGJ-707, positioned the product as a leading candidate in the evolution of immunotherapy. This collaboration and the continued investment into clinical trials are paving the way for more robust evidence and wider global acceptance of this innovative treatment.

The Phase III studies that follow will be critical, not just for 3SBio but for the entire field of lung cancer therapies. Success in these trials could redefine treatment protocols and lead to improved outcomes for patients worldwide.

As the cancer treatment community eagerly anticipates the next steps for SSGJ-707, the results presented at ESMO 2026 serve as a beacon of hope, signaling a potential shift in therapeutic approaches against one of the most arduous diseases.
