Innovent Biologics and Spero Therapeutics Forge Exclusive Agreement for Revolutionary Anti-CD40L Antibody

Innovent Biologics, Inc. and Spero Therapeutics, Inc. have finalized a significant exclusive licensing agreement for the development of IBI355, also identified as SP001. This third-generation anti-CD40L antibody is currently prepared for Phase 2 clinical trials, marking a pivotal moment in the treatment of autoimmune diseases such as IgG4-related disease (IgG4-RD) and Sjögren's disease (SjD).

Under the terms of this agreement, Spero will secure exclusive global rights, with the exception of Greater China, to research, develop, manufacture, and commercialize IBI355. In exchange, Innovent will receive an initial financial payment and has the opportunity to earn development, regulatory, and commercial milestone payments, totaling approximately $1.1 billion. Additionally, Innovent is set to receive tiered royalties on net sales in the territories where Spero holds the license.

IBI355 is regarded as a key advancement in the treatment of autoimmune conditions due to its targeting of CD40L, a critical immune signal protein that influences multiple immune pathways. Spero Therapeutics anticipates launching a Phase 2 clinical trial for IBI355 in patients with IgG4-RD in the second quarter of 2027. Likewise, Innovent plans to commence a Phase 2 trial in China targeting SjD, which is a widespread autoimmune disorder that predominantly afflicts women and is associated with severe symptomatic burden.

According to Dr. Arezou Khosroshahi from Emory University, interrupting CD40L signaling may represent a crucial area for clinical exploration in IgG4-RD, which suffers from a lack of treatment options that adequately address chronic inflammation and the need for effective management of relapses.
