Since the implementation of USP <382> on December 1, 2025, producers are required to evaluate the functional suitability of elastomeric components not merely as standalone items but as integral pieces of the overall packaging or delivery system. This shift necessitates a comprehensive assessment of the elastomer’s performance capabilities, particularly regarding its ability to maintain container closure integrity.

The joint session, aptly titled "Are You Ready for USP <382>? A Complete Approach to Elastomer Resealing Capacity and Container Closure Integrity," will feature insights from Dr. Francesca Caprioli, Laboratory Director at LIGHTHOUSE, and Brandon Zurawlow, Chief Scientific Officer at CS Analytical.

As the conversation around elastomeric performance expands, it now includes a broader spectrum of evaluations related to package integrity and the functional aspects associated with drug delivery systems. Brandon Zurawlow emphasizes, “USP <382> moves the conversation around functional performance of elastomers beyond pharmaceutical vials, including prefilled syringes and cartridges, making the evaluation process more comprehensive.”

The webinar promises to elucidate the key modifications introduced in USP <382> compared to its predecessor USP <381>, particularly the transition from traditional blue dye tests for needle self-sealing evaluations to utilizing various container closure integrity methods referenced in USP <1207>. Additionally, this shift will spotlight inherent integrity evaluations that must now be incorporated into the testing protocols.

Attendees can look forward to in-depth discussions on the application of laser-based headspace gas analysis, allowing for the detection of container closure integrity through alterations in headspace pressure or gas concentrations. Highlights will include:
  • - Real-time monitoring of elastomer resealing behaviors after multiple needle punctures under different conditions, including nitrogen and vacuum environments.
  • - The innovative carbon dioxide tracer gas ingress method that can uncover defects as small as 5 μm.
  • - How to integrate various testing methodologies to create a robust USP <382> compliance strategy.
