Context of the Study


The skin’s barrier function has traditionally been viewed as primarily due to the stratum corneum, the outermost layer of the epidermis. This layer is composed of a lipids matrix, dominated by ceramides, which creates a lamellar structure critical for moisture retention and protection against external irritants. However, recent findings have illuminated the essential role of the tight junctions located below the stratum corneum, which are critical in safeguarding against foreign substances and irritants by acting as a seal between cells.

Despite the acknowledged importance of tight junctions, there has been limited exploration of cosmetic ingredients that can bolster or protect this function. This gap in research prompted Rosette Co., Ltd., in collaboration with Utsunomiya University, to investigate the roles that natural ceramides might play beyond the traditional scope of skin barrier enhancement.

Key Findings of the Research


Utilizing human epidermal cells for analysis, the researchers discovered that natural ceramides significantly increase the expression of tight junction-related molecules such as Claudin-1 and Occludin. This increase correlates with enhanced tight junction barrier function. Importantly, the study also found that natural ceramides could inhibit the impairment of tight junction functionality due to ultraviolet (UV) radiation. This reveals that natural ceramides act not only on the stratum corneum barrier but also represent a class of bioactive lipids that enhance the secondary skin barrier, the tight junctions.

Expert Insights


Professor Genji Imokawa from Utsunomiya University highlighted, "Natural ceramides have been recognized for their role in increasing stratum corneum ceramides to improve skin barriers. Our research suggests they may also affect tight junctions, the second skin barrier. This is significant as it indicates that natural ceramides could simultaneously support both barrier functions."

Future Implications


The implication of this research is profound, particularly in relation to conditions like atopic dermatitis and sensitive skin, which not only show compromised stratum corneum barriers but also weakened tight junction functions. The findings open new avenues for sensitive skin care, atopic dermatitis skincare regimens, protection against UV damage, and countering barrier impairment due to aging.

More than just traditional hydration, this research proposes a novel skincare strategy focused on re-establishing the skin’s barrier mechanisms. Rosette intends to further explore the efficacy of natural ceramides in enhancing skin health and will utilize these insights to spearhead product development aimed at improving skin barrier function.
