In a significant advancement in vascular therapy, Advanced NanoTherapies, Inc. (ANT) has successfully raised over $31 million in a Series B financing round. Redefining Drug-Coated Balloons


Advanced NanoTherapies is charting a new course in the realm of drug-coated balloon (DCB) technologies with the introduction of SirPlux Duo. This device innovatively incorporates a proprietary nanoparticle delivery system that enhances cellular uptake within blood vessel walls. By combining the antiproliferative effects of two well-established drugs, sirolimus and paclitaxel, SirPlux Duo aims to provide a more effective treatment than traditional single-drug coats.

"This financing milestone arrives at a pivotal moment in vascular intervention. While first-generation single-drug DCBs are gaining traction in the U.S. market, they expose the limitations of passive crystalline drug delivery," stated Marwan Berrada-Sounni, CEO of ANT. Addressing Clinical Challenges


Restenosis after percutaneous coronary intervention (PCI) remains a persistent clinical issue, predominantly driven by cellular proliferation at treatment sites. The new platform is designed to administer both drugs simultaneously to the lesion site utilizing functionalized nanoparticles. This system not only ensures sustained local drug retention but also achieves higher efficacy while using lower doses.

Dr. Rishi Puri, an interventional cardiologist from Cleveland Clinic, echoed the potential impact of this technology: "SirPlux Duo promises to enhance outcomes by utilizing dual-drug delivery with controlled cellular uptake and improved tissue retention. This leap forward in vascular intervention could lead to outcomes comparable to those achieved with drug-eluting stents while adhering to the intervention principle of leaving nothing behind."

Clinical Evidence and Future Steps


The technology behind SirPlux Duo, exclusively licensed from the Cleveland Clinic, is currently undergoing clinical evaluation in the ADVANCE-DUO study. The initial data has been promising, with a first-in-human trial involving 28 patients across Australia, New Zealand, and the Dominican Republic demonstrating no new treatment failures after a two-year follow-up period. Furthermore, ongoing studies in Spain aim to replicate the U.S. pivotal protocol, promising rich data for future regulatory submissions.

Moving forward, Advanced NanoTherapies plans to scale up manufacturing in compliance with FDA IDE requirements and will continue progressing towards pivotal execution for both coronary indications and below-the-knee peripheral applications. Their roadmap includes IDE submissions for in-stent restenosis and small-vessel de novo lesions and pursuing pivotal clinical trial agreements with the FDA.
