Paradigm Therapeutics Advances Phase 3 Trial for Innovative EB Treatment, SD-101 (Zorblesia™)

Paradigm Therapeutics Initiates Phase 3 Trial of SD-101 (Zorblesia™)

Paradigm Therapeutics, Inc., a biopharmaceutical company specializing in innovative therapies for rare diseases, recently announced the enrollment of its first patient in the Phase 3 clinical trial for its investigational treatment, SD-101, also known as Zorblesia™. This topical cream aims to address chronic wounds and skin lesions associated with Epidermolysis Bullosa (EB), a debilitating genetic disorder.

Understanding Epidermolysis Bullosa (EB)

EB is a group of rare genetic disorders marked by extremely fragile skin that is prone to blistering and tearing with minimal friction. Symptoms can appear at birth or in early childhood and may persist throughout an individual's life. The condition often leads to chronic wounds, significant pain, persistent itching, infections, and scarring, creating a considerable burden for patients and their families. Moreover, the severity and subtype of EB can vary, leading to varying degrees of mobility and quality of life.

The Phase 3 Study: Objectives and Protocol

The clinical trial, referred to as SD-007, is a multicenter, randomized, double-blind, placebo-controlled study aimed at evaluating the efficacy and safety of SD-101 across three major EB subtypes: EB simplex (EBS), recessive dystrophic EB (RDEB), and junctional EB (JEB). In this study, approximately 80 participants, aged between 28 days and 12 years, will be recruited from 17 clinical sites distributed over 14 countries, which include regions in North America, South America, Europe, Asia, and Australia.

Participants in the trial will apply SD-101 or a comparator cream daily over their entire body for a duration of two months. The primary focus of the study will be to assess the closure rate of targeted chronic wounds, evaluate changes in the body surface area affected by wounds and lesions, and monitor improvements in itching symptoms.

Paradigm's Commitment to EB Patients

Dr. Robert Ryan, President and CEO of Paradigm Therapeutics, expressed his belief that there is a pressing need for effective treatments for individuals with EB, noting the limitations of current therapeutic options. "SD-101 has the potential to transform how wounds and lesions are treated across the skin's surface in patients suffering from EB simplex, RDEB, and JEB," he stated. Initiating the Phase 3 study is perceived as a significant step forward in the development of this innovative therapy.

Previously completed Phase II and III studies have indicated promising results, highlighting SD-101’s potential to aid in wound closure and lessen the overall skin area impacted by lesions. Paradigm is focused on bringing SD-101 closer to regulatory approval, aiming to provide new treatment options for patients battling this challenging disorder. Furthermore, patients finishing the SD-007 trial may also have the opportunity to continue their treatment in a subsequent study, SD-008.

Community Support and Future Implications

Dr. Fred Eshelman of Eshelman Ventures, an investor in Paradigm, emphasized the urgency of expanding treatment options for EB patients and applauded Paradigm's efforts in advancing SD-101. The company is diligently working with potential partners to support the global development and accessibility of this promising new treatment.

Conclusion

With SD-101 (Zorblesia™) backed by Rare Pediatric Disease and Orphan Drug designations, Paradigm Therapeutics is paving the way for innovative therapies that could significantly enhance the lives of countless individuals affected by Epidermolysis Bullosa. As the clinical trial progresses, many are hopeful that this groundbreaking approach will fulfill a critical unmet medical need, offering new hope to patients and their families facing the challenges posed by this relentless condition.
