IDEAYA Biosciences and Servier Present Promising Darovasertib Data for Uveal Melanoma at ESMO 2026
IDEAYA Biosciences and Servier Present Groundbreaking Data on Darovasertib for Uveal Melanoma
In a significant advancement for cancer therapy, IDEAYA Biosciences, in collaboration with Servier, unveiled pivotal findings from three clinical trials assessing darovasertib during the European Society for Medical Oncology (ESMO) Congress 2026 held in Madrid, Spain. This new therapy demonstrates promise in treating both metastatic and primary uveal melanoma, emphasizing its potential to transform care pathways for affected patients.
Uveal melanoma, a rare and aggressive form of cancer originating from eye melanocytes, poses significant treatment challenges, particularly in managing disease progression. The data presented from trials OptimUM-01, OptimUM-02, and OptimUM-09 reveals darovasertib's potential as a foundational therapy across various treatment stages.
OptimUM-01 and OptimUM-02 Trials
OptimUM-01 and OptimUM-02 trials focused on assessing the efficacy of darovasertib in combination with crizotinib for patients diagnosed with metastatic uveal melanoma. Data gathered from OptimUM-01 highlighted that the synergistic approach offered substantial clinical benefits, irrespective of patients’ HLA-A0201 allele status. For treatment-naïve patients, the median progression-free survival (PFS) was remarkable at 7.4 months, while overall survival (OS) reached an impressive 27.1 months.
Among participants who were previously treated with tebentafusp, significant responses were also noted. With an overall response rate (ORR) of 22.1%, the results vastly outperformed historical benchmarks for metastatic uveal melanoma, characterized by a typical median PFS of merely 2-3 months.
OptimUM-02 trial data revealed consistent benefits for patients with HLA-A0201-negative status. This subgroup analysis showed marked improvements in ORR and PFS compared to investigator's choice therapies, reinforcing the proposition that darovasertib could set a new standard in first-line treatments for this challenging patient population.
OptimUM-09 for Neoadjuvant Treatment
Examining the role of darovasertib earlier in treatment, the OptimUM-09 trial investigated its use as a neoadjuvant therapy in patients requiring local treatment for primary uveal melanoma. This trial yielded promising figures with over 80% of patients experiencing tumor shrinkage, a critical milestone for advancing treatment approaches where eye preservation is critical. Notably, there was a 57% rate of eye preservation in patients undergoing enucleation, a finding that could significantly impact patients’ quality of life.
Dr. Yujiro S. Hata, CEO of IDEAYA Biosciences, expressed optimism regarding these outcomes, stating, "These updates underscore darovasertib’s ability to address significant unmet medical needs across the uveal melanoma treatment spectrum. We hope to shift the treatment paradigm by making darovasertib a go-to therapeutic option for both metastatic and primary cases."
Future Prospects
Building on this rich dataset, IDEAYA plans to submit a New Drug Application (NDA) to the U.S. Food and Drug Administration (FDA) for darovasertib in combination with crizotinib in the coming months, as part of their strategic effort to enhance access to this innovative treatment. The company aims to secure a role for darovasertib not just as a treatment option but as a critical component of uveal melanoma therapy as they navigate FDA review and further global regulatory pathways.
With ongoing collaboration with Servier, IDEAYA is positioned to expand further into the oncological landscape, promising hopeful advancements for patients diagnosed with uveal melanoma and potentially altering the course of treatment for this aggressive cancer.
The ESMO Congress 2026 will serve as a pivotal moment, showcasing these findings to a global audience of medical professionals and resourcing them to push forward the agenda of innovative cancer therapies.
All presentations at ESMO are scheduled for October 24, 2026, highlighting darovasertib’s exciting potential in clinical oncology, particularly for patients battling uveal melanoma.
