AbbVie Reveals Promising Phase 3 Results for Lutikizumab in Treating Hidradenitis Suppurativa
AbbVie Announces Promising Results for Lutikizumab in Hidradenitis Suppurativa
AbbVie has recently shared the topline outcomes from its Phase 3 trial, INTREPID, showcasing the efficacy of Lutikizumab, a new investigational treatment for patients suffering from moderate to severe hidradenitis suppurativa (HS). This study not only met its primary endpoint but also exceeded expectations by significantly improving multiple secondary endpoints, reflecting positively on its potential as a viable treatment option.
The INTREPID study established Lutikizumab's effectiveness by demonstrating a notable improvement in the Hidradenitis Suppurativa Clinical Response 75 (HiSCR75) at Week 16. Specifically, 31.4% of patients treated with Lutikizumab achieved this benchmark compared to only 23.4% of patients in the placebo group, marking a statistically significant difference. HiSCR75 represents a 75% reduction from baseline in abscess and inflammatory nodule counts, with no increase in abscess or draining fistula counts.
Further emphasizing the drug's efficacy, Lutikizumab showed advancements in eight ranked secondary endpoints, primarily related to pain management and flare reduction. Notably, at Week 8, a significant reduction in HS-related skin pain was reported. Among Lutikizumab recipients, 26.2% experienced at least a 3-point reduction on the Numeric Rating Scale (NRS), compared to only 15.0% for the placebo. The results highlight Lutikizumab's early impact as improvements were noted as soon as Week 1.
Patients treated with Lutikizumab enjoyed not only fewer flares of HS but also enhancements in their overall quality of life, as outlined by improvements in the Dermatology Life Quality Index (DLQI) and the Hidradenitis Suppurativa Impact Assessment (HSIA). The study also measured the reduction of inflammatory markers linked to HS pathogenesis, such as IL-17A and IL-17F, reinforcing the drug’s therapeutic promise.
From a safety perspective, Lutikizumab was well tolerated among participants. The occurrence of treatment-emergent adverse events remained largely similar between the Lutikizumab and placebo groups. The most frequently cited adverse events included nasopharyngitis, upper respiratory tract infections, headache, and fatigue. Notably, there were no new safety concerns raised during the trial, and the overall safety profile aligned with prior studies, with 53.4% of patients experiencing adverse events in the Lutikizumab cohort, compared to 53.7% in the placebo group.
Doctors have praised these findings, particularly given HS's complexity as a chronic, painful condition that persists despite available treatments. Dr. Alexa B. Kimball, President, and CEO of Harvard Medical Faculty Physicians at Beth Israel Deaconess Medical Center, commented, “The INTREPID findings are significant as they establish a rigorous primary endpoint while evaluating the dual inhibition of IL-1α and IL-1β. These outcomes support the potential of Lutikizumab as a valuable treatment option.”
Moving forward, AbbVie aims to present more detailed findings from the INTREPID study at upcoming medical congresses and anticipates further data from a 36-week extension period in 2027, which will support regulatory submissions for Lutikizumab. The company remains committed to improving the treatment landscape for HS, and its ongoing studies will explore combinations of Lutikizumab with other therapies to tackle various disease mechanisms.
With over 1% of the global population affected by HS and often facing long diagnostic delays, advancements like Lutikizumab might finally offer hope to those battling this debilitating condition. AbbVie’s dedication towards innovative solutions in immunology continues to pave the way for transformative change in the management of hidradenitis suppurativa and other challenging immune-mediated disorders.
About Hidradenitis Suppurativa
Hidradenitis suppurativa is characterized by painful skin lesions, cysts, and recurrent inflammation. Despite recent treatment advancements, many individuals with HS experience inadequate relief, making the development of effective therapies such as Lutikizumab crucial.
