Key Findings from the Trial


The twelve-month analysis indicated that participants receiving SKY-0515 showed a slight improvement in the TFC score compared to the expected deterioration based on historical control data. Specifically, the mean change for the TFC score was +0.07, which contrasts with an anticipated decline of -0.87, highlighting the potential of SKY-0515 in enhancing functional capabilities.

In terms of motor functions, the TMS score revealed a mean change of -2.00 for those on SKY-0515, significantly smaller than the expected worsening of 2.21 points. Cognitive assessments demonstrated a stabilization of the SDMT score at -0.19, far better than the predicted decline of -1.78 points, and the SWRT score saw an impressive gain of +3.44, while a decline of -3.13 was forecasted.

Furthermore, subjective evaluations from both clinicians and patients highlighted impactful results. Approximately 65% of participants and 50% of clinicians reported observed improvements rather than disease progression over the treatment duration, a substantial finding given the nature of Huntington's disease.

Mechanism of Action and Safety Profile


CRucial to the efficacy of SKY-0515 is its impressive ability to significantly reduce the levels of mutant huntingtin (mHTT), a protein directly linked to the disease pathology, by up to 69%. It also effectively lowered PMS1 mRNA levels, a significant contributor to the progressive nature of Huntington's disease, by about 26%. The therapy demonstrated excellent tolerability across a diverse participant pool, with over 175 individuals enrolled across five different countries.
