Documents reveal that Osaka City did not conduct its own verification regarding claims that blue mold produced by the company’s factory was responsible for generating Puberulic acid, leading to contamination in their red yeast rice product.

On August 18, 2023, a request for clarification was sent to Osaka City's Health Bureau, seeking information on how they determined that the species of blue mold identified (namely Penicillium adametzioides) was capable of producing Puberulic acid. The deadline for a response was set for August 21, but no reply was received by that date.

The questions posed were straightforward: what scientific evidence exists to support the claim that the identified species constitutes a strain that produces Puberulic acid? Identification based solely on species designation is insufficient to confirm the strain's ability to generate specific metabolites. Analyzing not just the species but also the genetic characteristics and culturing conditions is essential for such verification.

In earlier responses dated July 18, the Osaka City Health Bureau acknowledged that genetic testing can only estimate species identification. Furthermore, they noted that the strain IFM 68223, used for verification, was not standard and was obtained to assess the production ability in other strains of P. adametzioides. Thus, when questioned about the production capabilities of the strain derived from Kobayashi’s factory, the Health Bureau failed to supply any evidence or basis for their claims regarding its ability to produce Puberulic acid.

The Health Bureau explicitly admitted that it did not independently verify the claims made by Kobayashi Pharmaceutical. In a separate document dated August 17, it was stated that the information regarding the contamination was derived from Kobayashi's own checks, with no records of the city independently verifying those findings or the decision not to conduct such verification.

Concisely, key insights drawn from this situation are as follows:
1. The Osaka City Health Bureau did not address questions regarding the confusion between species and strains.
2. They acknowledged that they did not verify the strains in question.
3. No scientific basis was offered linking the presence of the P. adametzioides species in the factory to the strain’s alleged ability to produce Puberulic acid.

In light of the above points, documents obtained indicate that the Osakan authorities did not confirm any scientific basis for the assertion that the factory-derived strain was indeed capable of Puberulic acid production. Therefore, the claim that the blue mold from the factory produced Puberulic acid, which subsequently contaminated the red yeast rice, lacks substantiation as per the currently available public documents from Osaka City.
