Sino Biological's Cell-Free Protein Synthesis Transforms AI Protein Design Validation in Recent Study

Sino Biological's Innovative Approach to Protein Design

Sino Biological, Inc., a leading player in genetic synthesis and protein production, has made waves with its cell-free protein synthesis workflow. This groundbreaking technology is highlighted in a recent Protein Design study published in Nature Communications. The research, which emerged from the Tencent AI for Life Sciences Lab, showcases a new method that allows for the swift validation of AI-developed proteins, boasting enhanced activity, stability, and multifunctionality.

Bridging AI Protein Design with Experimental Validation

Artificial intelligence has accelerated the design of protein amino acid sequences, but translating these computer-generated designs into functional proteins remains a significant challenge. The complexity of protein activity, stability, folding, and expression is influenced by numerous structural and biochemical factors, often leading to discrepancies between in-silico predictions and experimental outcomes.

To bridge this gap, the study introduced an Ontology Reinforcement Iteration (ORI) framework, which combines protein ontology with reinforcement learning derived from wet-lab feedback. This iterative model continuously refines the protein sequences based on experimental data, including protein expression levels and functional activities, thus improving design accuracy.

Accelerating the AI Design Cycle with Cell-Free Synthesis

The researchers utilized the XPressMAX™ Cell-Free Protein Synthesis Kit from Sino Biological, which enabled rapid protein expression and functional screening. By incorporating protein-coding sequences into the kit's expression vector and adding them to the proprietary cell-free reaction system, the study supported fast design-build-test cycles. This innovative workflow led to the development of a lysozyme that exhibited more than 100 times the activity of its natural counterpart, a thermostable chitinase that retains activity at 85°C, and bifunctional enzymes that outperform naturally occurring multifunctional enzymes.

Key Features of the XPressMAX™ Kit

The XPressMAX™ Kit not only streamlines the protein synthesis process but also offers several advantages:
  • - Ultra-Fast Protein Synthesis: Proteins can be expressed in as little as 3 hours.
  • - High Screening Efficiency: Optimized for VHH, scFv, and miniproteins.
  • - Flexible Template Support: Compatible with plasmid and linear DNA templates.
  • - Disulfide Bond-Friendly: Facilitates the expression of complex disulfide-bonded proteins without additional enhancers.
  • - Cost-Effective Performance: Reduces operational time and costs significantly.
  • - Scalable System: Suitable for high-throughput and industrial applications.
