Kazia Therapeutics Unveils Promising Preclinical Results for Paxalisib Against Colorectal Cancer

Kazia Therapeutics, a Sydney-based biotechnology firm listed on NASDAQ under the symbol KZIA, has recently released compelling preclinical data concerning its anti-cancer drug, Paxalisib. This innovative compound has demonstrated remarkable efficacy in reducing tumor burdens in microsatellite stable (MSS) and proficient mismatch repair (pMMR) colorectal cancer models, specifically highlighting its ability to enhance responses to existing immun therapies. In one study, Paxalisib alone yielded a significant 52% reduction in tumor volume (p=0.035), while when combined with the established immunotherapy pembrolizumab, it resulted in an additional 50% decrease (p=0.022).

The research further indicates that Paxalisib was well tolerated by participants, showing no signs of treatment-related toxicity. This is particularly encouraging given the urgent need for effective treatments against colorectal cancer, which is a leading cause of cancer-related mortality worldwide. Much of this challenge lies in the resistance often observed with current immunotherapies in treating colorectal cancer, especially among the 85-90% of patients with MSS/pMMR type. Existing studies have shown that, unlike their microsatellite instability high (MSI-H) counterparts, the MSS/pMMR tumors do not respond well to checkpoint inhibitors, making Paxalisib's approach potentially groundbreaking.

Kazia Therapeutics is poised to further investigate Paxalisib's capabilities with plans for a Phase 2 clinical trial that includes a five-arm study. This will evaluate Paxalisib, both as a standalone treatment and in combination with pembrolizumab, against standard care for pre-treated MSS/pMMR metastatic colorectal cancer. The study will prioritize assessing safety and tolerability while also examining progression-free survival, overall response rates, and overall survival as secondary endpoints. Patient enrollment is anticipated to begin in early 2027, with the aim of completing recruitment by the end of the same year.

The innovative research suggests that Paxalisib may effectively reprogram the tumor microenvironment, thereby enhancing the visibility of cancer cells to the immune system. In addition to Paxalisib, Kazia is actively advancing various clinical programs targeting other forms of cancer. This includes a next-generation biomarker program designed to elucidate the molecular and immune characteristics associated with responses to Paxalisib, allowing for refined patient selection and individual treatment strategies. This program is expected to improve predictions of treatment outcomes and develop tailored biomarker strategies critical for the drug development process.
