Major Investment Boosts Research for Neurodegenerative Diseases Linked to Tau Proteins

Groundbreaking Research Initiated for Tau-Driven Diseases



In a significant move to combat neurodegenerative diseases associated with tau protein accumulation, the Rainwater Charitable Foundation has announced an impressive $3 million investment to fund four pioneering research projects. These awards are part of a broader initiative aimed at unraveling the complexities surrounding tauopathy—a group of debilitating conditions, including Alzheimer's disease, Progressive Supranuclear Palsy (PSP), and Corticobasal Degeneration (CBD).

The announcement, made in collaboration with key partners such as the Alzheimer’s Association®, the Aging Mind Foundation, and CurePSP, marks a notable step in addressing the urgent need for innovative therapies in a field characterized by limited treatment options. Despite advances in understanding the biological mechanisms of tauopathies, many questions remain regarding how these proteins lead to neurodegeneration.

Each of the selected researchers will receive $750,000 to explore various aspects of tau biology over the next two years. These projects were selected during the recent Tauopathy Challenge Workshop, emphasizing collaborative efforts to challenge the norms of neurodegenerative disease research.

The Research Projects and Their Goals


1. Carlo Condello, PhD (University of California, San Francisco)

Condello aims to investigate the molecular diversity of tau proteins specific to cell types in 4-repeat tauopathies such as PSP and CBD. By utilizing human brain samples and advanced stem cell models alongside animal studies, the research seeks to demonstrate how different cellular environments influence tau’s structure and the formation of disease-associated strains. This investigation may pave the way for developing targeted therapies that address the unique characteristics of each neurodegenerative condition.

2. Daniel C. Lee, PhD (University of Kentucky)

This project will delve into the relationship between citrullination and phosphorylation in modifying tau's behavior. Unlike phosphorylation, which is reversible, citrullination creates a permanent change to tau, thus potentially leading to distinct pathological forms. Lee's work aims to quantify how this irreversible modification impacts the aggregation and structural behavior of tau, providing fresh perspectives on protein misfolding, crucial to understanding tau-driven diseases.

3. Edward B. Lee, MD, PhD (University of Pennsylvania)

In his research, Lee will focus on the bundling of tau fibers within neurodegenerative conditions like PSP, along with their interactions with nearby cell structures. Employing cutting-edge techniques such as cryo-electron tomography, he hopes to identify why certain brain regions are more affected than others and how variations in symptoms can be explained through differences in tau pathology across different patients. This insight could lead to targeted interventions and improved therapeutic options.

4. Sue-Ann Mok, PhD (University of Alberta)

Mok’s work seeks to explore tau aggregation more comprehensively through innovative techniques like limited proteolysis assays and single molecule mass photometry. By studying the early aggregation reactions of tau, her research aims to identify specific modifications that could either enhance or inhibit the formation of disease-specific aggregates. This research could be instrumental in developing diagnostic and treatment strategies based on the tau aggregation mechanisms.

As a leading independent funder of neurodegenerative research, the Rainwater Charitable Foundation has committed over $165 million to advancing our understanding of these complex diseases. The foundation's President, Jeremy Smith, emphasized, “Investing in innovative science is essential for unlocking new paths toward diagnosis, treatment, and prevention in the face of growing neurodegenerative burdens.”
