This innovative strategy aims to supplement existing intramuscular respiratory vaccines with a mucosal booster, thereby enhancing the immune response at the mucosal level where respiratory pathogens first enter the body.

Current intramuscular vaccines are known for their ability to generate strong systemic immunity, effectively protecting individuals from severe disease. However, they often provide limited mucosal immunity, a crucial factor in reducing transmission at the site of entry. AdJane posits that by integrating mucosal components into existing vaccination regimens, the ability to mitigate both disease severity and transmission can be improved significantly.

The Prime-Boost Approach


AdJane's strategy revolves around a prime-boost method, which employs an initial intramuscular vaccination followed by a mucosal boost via a nasal spray using Nm-nOMV technology. It highlighted that participants who had previously received mRNA vaccines (like those from Pfizer or Moderna) responded well to subsequent intranasal vaccinations with Nm-nOMV expressing the SARS-CoV-2 spike protein. This response was characterized by significant levels of IgA antibodies in the nasal mucosa, which are crucial for mucosal immunity.
